Rookie Officer Cooper (Witherspoon) is tasked with escorting Daniella Riva (Vergara) the wife of a dead drug dealer, to Dallas as part of a witness protection program. Things go wrong during the journey with Cooper not knowing who to trust as the line between the good cops and dirty cops are blurred.
Vergara's Daniella is every cliche and stereotype of a bombshell Latina all rolled into one dress-bursting, outrageously sexy body. Reese's Officer Cooper cops a superior attitude and talks like she ate up a police officer's rule book when she was little. And speaking of little, the diminutive Cooper is only 5' 2" while the well-built Daniella towers over her. Physically, mentally and emotionally, they couldn't be more different. And they are lumped together for the ride from San Antonio to Dallas with Daniella trying every chance to escape as Cooper struggles to get her to Dallas the next morning by hook or by crook. Daniella's husband had decided to testify against kingpin Vincente Cortez (Cosio), but is shot by gang members before he can do so. He leaves behind a distraught Daniella who spouts the kind of cuss words that would make a sailor blush.
Cooper and Riva escape the shooting scene but Cooper is soon shocked when two cops she trusts are actually on Cortez's payroll. They're on their own and the road to Dallas also sees Cooper coming into her own as a seasoned cop. It's also easy to tell that by the end of the movie, Cooper and Daniella will become besties.
It's a good thing this movie makes no remote hint of a pretense to take itself seriously because going by the gag reel at the end credits itself (yes, stick around for that) it's clear that the cast members themselves had more than a bunch of laugh-out-loud moments while shooting. The movie's humour rides on the random pairing of these two ladies.
Hot Pursuit is a film that takes the whole buddy-movie formula and gives it a wild, sometimes boisterous and genuinely funny female spin.
Shooting was done extensively in real locations, including grocery stores in small town America, including New Orleans, Louisiana.

We see Cooper being brought up by her father, a police officer, who had her travelling in his cop car almost wherever he went. Through the years, she learns all kinds of police lingo including all the codes to report various illegal incidents. Ithe n the present day, we see her as a cop who is assigned to fairly menial duty (read: a glorified secretary as one of the cops calls her) in her cop station. One day, her boss gives her the opportunity to get out there and prove her mettle. The assignment is a dangerous one. She is asked by her boss Captain Emmett (Lynch) to help transport a witness to testify in Dallas from San Antonio. At the cop station, Cooper is introduced to Detective Jackson, who is a towering presence in the police department and a superior officer go to the witness' residence in the countryside and collect Felipe Riva (Vincent Larisca) and his wife Daniella and head out. Daniella delays getting ready and as Cooper goes upstairs to her bedroom to find out why she is taking so long, two pairs of gunmen break in and in the ensuing firefight, both Jackson and Felipe are killed. Cooper and Daniella manage to flee the house and later, Cooper contacts her fellow police officers, Detective Dixon (Mosley) and Detective Hauser (del Negro) for help, she discovers that they knew that she would be out there. After glancing at their car's fuel gauge, she does some quick math and calculates that if the two detectives had driven from the police station to where they are, they wouldn't still have that much fuel still left in the tank. Cooper realizes that the two Detectives are crooked and are on the drug lord Cortez' payroll and that that they will kill the two of them. Cooper and Daniella make an excuse to visit the restroom, saying that the latter has got her chums and needs to change her tampon. They then escape and realize that the both of them are now considered wanted fugitives. More precisely, Cooper is being projected as armed and dangerous - a renegade cop. They go to a store and change their clothes, ditching their old and easily identifiable outfits.
There are a series of funny sequences, such as them trying to rob a farmer's truck and pretending to be lesbians to convince the farmer that they are just simply lost. They also manage to steal a pickup truck, which belongs to Randy (Kazinsky), who helps them find shelter in a casino hotel. They have a fight in their room and somehow manage over time, to work things out. Cooper asks Daniella why she is packing so many shoes and the latter tells Cooper that they were gifts from her brother, who was killed by the same cops he tried to seek help from, when targeted by the drug mafia in Colombia. This is why, she explains, Daniella hates cops. Later they get on a bus for Dallas and are again tailed by Detectives Hauser and Dixon but those two are blown off the road by two gunmen.
It appears that these two are in Daniella's employ. Cooper then realizes that Daniella had got her husband knocked off and was now going to go after Cortez' daughter and Cortez himself as the latter had killed her brother - the only family she had. Daniella plans to do this by attending a function for Cortez' daughter. Cooper knows of this and against the advice of Captain Emmett, also attends dressed first as a boy and then, as one of the serving ladies. In the final showdown, Cooper manages to convince Daniella not to shoot Cortez as she will be in jail for the rest of her life, as compared to a measly few months for her current charge of obstructing justice. As Daniella turns away, Cortez tries to shoot her but is in turn shot by Cooper. We cut three months into the future, Cooper picks up Daniella from jail after the completion of her term and Cooper has also got together with Randy. We see the three of them laughing and joking as they drive off together.
